With aim to maximize sensitivity and optimize LOD of this assay the volume of sample retrieved from the Cd column was maximized (150 mcrL) and reagent sandwich was assembled, as specified in the assay protocol (2.2.19). Should the goal of the assay be, to cover concentration range up to 1000 ppB N the sample volume retrieved from Cd column should be decreased to 20 mcrL (2.2.20.). Note that the protocol for the concentration ranges (A) and (B), shown here, was the same, the improved detection limit being due only to the scaling of the response of the spectrophotometer.
